The DHF2 gene encodes for dihydrofolate reductase that converts dihydrofolate into tetrahydrofolate, a methyl group shuttle required for the de novo synthesis of purines, thymidylic acid, and certain amino acids.

A deletion in this gene means a poor conversion of folic acid to tetrahydrofolate. This has led many genomists to favour supplementation of the bioavailable form of folic acid, methylated folate - 5,10 MTHF.
